The weight of the hot dried sample will be flactuating contineously due to the fact that the hot sample tends to absorption moisture from the environment very fast. So you must cool the hot dried sample into the desiccator before taking the final weight.

We can use a single sample, let's say 10 grams of liquid milk. Use IR or Oven method to determine the moisture in liquid milk. By subtracting the moisture weight, we'll get total solids weight. Use total solids weight to run the soxhlet extraction and determine the fat content. Finally we'll also get the non-fat solids. I guess this way is more faster and convenient to get all 4 components from a single sample. What do you think?

I have two questions the first one is that, is this procedure can be applied similar to a liquid, semi-liquid and volatile chemicals and the second one is, since temperature and time are inversely proportional, can we reduce the 2 hr drying to 1 hr by raising the temperature to 270'c? thank you!
You can use this method for liquid and semi solid samples but not for volatile substance. Because, at high temperature sample will be removed with the removal of moisture. But if your test parameter is "Moisture & Volatile Substance", then you can follow this method for testing a volatile substance. Secondly, you can not use temperature higher than 133 degree Celsius. Otherwise the sample will burn out slightly, therefore loss of sample.
I have a question. Do i need to dried mushroom first for measuring Dry weight moisture content of mushroom. Or i should directly measure the fresh mushrooms. Fresh mushrooms contains around 80-90% moisture, but for the analysis of nutrient content, this is not possible. So should i take the dry matter or dried it first
It depends on what you want. If you want to determine moisture of fresh mushrooms them go for it without drying. But if you want to dry then you must keep a note on the test result that the analysis was done after drying (on dry basis)

Nur Solehah, this is not the ideal method to determine the moisture content of a chemical sample. But you can analyze chemicals having boiling point higher than 200 degree Celsius according to this video because volatile chemicals evaporates before the moisture evaporation.

Naa Kadir, You can determine the moisture content of a solid sample following this method but you have to grind your sample before taking it for testing. Sample Grinding should be done as fast as possible to avoid extra moisture absorption from the environment by the sample.

Absorption of moisture by the sample from the environment may give questionable test results. Thats why we should weigh sample fast as much as possible.
If your test parameter is "Moisture & Volatile Matters" then you will get the actual result. But if the test parameter is "Moisture Content" then this method is not applicable for the sample which contains volatile matters.
