      This is a playlist i found on module 6 it should help ,but Biorach goes into much more detail and depth than these but they are still good to use.

    After 3. Is the temperature again raised to 95 degrees frm 72 degrees??

    When the free nucleotides pair up, does the primers remain bound? Or do they leave allowing free nucelotides to bind to the part of DNA it has left exposed?

      As far as I know the primers stay bound, but I'm not entirely sure... Sorry!

    second - i have a question about the third step (synthesis of DNA) - do the 2 strands join together by DNA polymerase or Taq Polymerase? (dna polymerase is whats mentioned in my text book)

      Heya i have the ocr textbook- it says DNA polymerase adds bases to the primer, and the enzyme Taq polymerase is used. I'm pretty sure what this means is that Taq polymerase is a type of DNA polymerase, which is used in PCR because it is able to withstand the extremely high temperatures and doesn't denature. Hope this helps :)

      Precisely! :) Taq polyermase *is* DNA polymerase found in the thermophile Thermus aquaticus. It is specifically isolated to use in PCR as it can withstand the high temperatures used in the thermocycler.
