There is no such thing as a mouse embryonic fibroblast. Fibroblasts don't exist in embryos. Your feeder layer is comprised of mouse fetal fibroblasts, typically derived from approximately 13.5 dpc murine fetuses.

Regarding the stem cells from URINE obtained after urination: HOW MANY TYPE OF STEM CELLS CAN WE FIND IN THAT URINE ? We can find only USCs ( urine stem cells ) or we can find also other stem cells ? . Is there any public protocol to separate if there are many types than USCs? Thank you.

The media is kinda yellow. Normally, I keep my media dark orange - red for Lymphocytes.
Is the growth conditions for ESC's different or is the media just light orange by formulation? Would adding more media disturb the cells too much?

According to Dr. Gamm, the color of the media depends on how dense and active the culture is. He conveyed that this might have been a particularly dense culture and not the best example.

What is temperature of the incubator? How are the cells stuck to the bottom of tray?

The answer to your second question is ''contact inhibition''.
