Getting a comfortable muzzle is definitely a significant part of muzzle conditioning. If the muzzle isn't comfortable then it doesn't matter how much positive association you've done, your dog won't like wearing the muzzle.
Unfortunately, my dog trainer said that the baskerville ultra muzzle I got (based on his sizing) was right and my dog just had to get used to it, but my dog has never been comfortable in it, and I wish I had started out with a well fitted muzzle.
