The Kingdom of Borgia is infamous for constantly experimenting with poisons. Cantarella was one of their most famous poisons, known to not only kill at precise timings depending on the amount administered, but also mimic the symptoms of a disease. Because of their constant use of poisons, there was a saying that went something like "Dine with the Borgias and you'll not come out alive." So back to the main point, even if Miku/Lucrezia seemed like the victim, she's also in love with her older brother Kaito/Cesare. Honestly, it took so much just to find out about this, but now that I did, it's kind of interesting to think about.
And I thought the title had something to do with singing... Not only because _"canto"_ means "singing" in Italian, but also because this is Vocaloid and all that...

This is the story in my perspective. Things went by in a blur and soon enough they found theirselves on the mattress. Kaito who was slowly taking his coat off was highly aroused, but upon Miku's soft and innocent sound, memories of their innocent and sweet childhood came rushing back, Kaito stopped and realized what he was just about to do to the person he loves and couldn't help but cry for the person he has turned into. He was so embarrassed by the fact and it was painful for him too. But just when he was about to leave, Miku who is actually conscious pulls him by his hand and whispers "Please, Capture me". The childhood couple's forlorn is reunited once more.

"OK, vocaloid history lesson. ^_^
Kaito and Miku are brother and sister in the video. To be more specific, they are expies of the infamous Borgia siblings--Kaito is Cesare Borgia; Miku is Lucrezia Borgia. (While Len bears most similarity with the figure of Alfonso de Aragon.)
Here are the details:
The whole story of Cantarella is loosely based on the story of the Borgias. In the video, Kaito and Miku are members of the "Vocalgia" family. They were very close as children--seeing themselves as two of a kind, but in time their feelings for each other grew into something which they knew can not be. This tormented both of them. Also, there are already plans for Miku to be married off to another powerful family. Something her own brother, Kaito, is painfully aware of. (What's more, since he's the one doing the conquering, he's the direct beneficiary of Miku's upcoming match as it would give him the power and the support he needs to conquer the land under his family name.) Kaito is an extremely ambitious but borderline tyrannical man who would do anything it takes to get what he wants; however, he has one soft spot: it turns out his love for power is only surpassed by his love for his sister. And when he sees the handsome and charming Len, he begins to grow increasingly jealous and insecure over the fact that Miku might actually come to like--even love--him in time. His heart grew dark. It got so bad he decided that he was going to be the first to take her--even if it means using his family's trademark poison--La Cantarella--to do it. But in the end he couldn't bring himself to do it. He desired Miku with all his heart, but he couldn't bring himself to "hurt" her. And decided to just lay bare his feelings for her. Luckily for him...Miku was well aware of his dark intentions and would have willingly went along with what he wanted to do even if he didn't choose to restrain himself in the last minute. It turns out Miku, was devious as he was, and reciprocated his forbidden love. She agreed to be his for one night, and it made them both very happy. Miku then assured Kaito that though she was married to another man, Kaito would remain her true love. With that Kaito was able to let Miku go to the altar, and she married Len without a hitch.
Unfortunately, all of this didn't last very long.
Kaito's resentment and jealousy never subsided and only grew as time went by--especially when he saw that Miku actually did find a measure of happiness with her husband. He couldn't take it anymore, and he decided to kill Len. Despite knowing full well, that doing so would damn his ambitions and bring the wrath of a powerful family upon his head. He didn't care anymore. He hated Len, and if getting rid of him meant having to fight against all the lords of the land at once--so be it. Kaito slipped some poison in Len's drink (or used some weakening drug, so that Len could be led outside and killed by Kaito personally). Miku...was confused. And horrified. She knew exactly what was going on, but the whole scenario was so twisted and cruel--so out of hand; she was torn between the man she loved all her life, and the man she had come to love in another way. The part of her that loved Kaito was also aware that what he did had grave consequences. She feared for his life...and in time...Kaito finally paid the price for his sins. He was killed in battle, presumably with the help of Len's family, and his dark ambitions were put to an end. The Vocalgia was powerful no more. Miku survived though. Her tenacity got her through in the end and after Kaito's downfall she either lived alone or married another man out of practicality, and judging from her clothes she continues to be well-off despite the loss of her family prestige. The years then passed...Miku matured, and she remained quite beautiful, but no matter how much time has passed she still couldn't forget the dark brother who loved her so madly and desperately--and whom she loved in turn. She visits his grave as often as she could, and is often haunted by his memory and his apparition--wanting so much to see and touch him again.
And, near the end? It's heavily implied that this is exactly what happens. After their earthly tragedy has ended--the forbidden lovers have reunited in death. 1507 is the year Cesare Borgia fell in battle--the year Miku "lost" Kaito; 1519 was the year Lucrezia Borgia passed away--the year Kaito "found/caught" Miku again."

I doubt you'll see how that song's lyrics are actually so horrible that it's beautiful in an artistic kind of way, but you will have a field day yelling "RAPERAPERAPERAPE". Note: Yes, rape is a bad thing, but it's just a song. A song pretty much made by someone to show how fucked up it is. Many people have done this. Kurousa-P (the author of this song) has made other messed up songs before (not really rapey, but still pretty messed up) aside from this one, such as "Acute". Hell, it's a song that actually tells a story.

Kaito gets to see Miku once again after many years of been separated, but thing is she is with someone else (Len), full of jealousy, Kaito goes after him and kills Len, then invites Miku for a drink and drugs her, yes he planned to take advantage of her, but then remembers his happy childhood with her, it is also revealed that they were force to be separated because Miku was already engaged to Len and he told on them when he saw them kissing, forcing the separation, back from the memory, Kaito finds himself unable to take advantage of her and Miku feels his tears understanding why, and she comforts him by admitting that she also loved him
