Description: Here, we describe a new animal model of cardiac toxicity following DNA damage that implicates cGAS/STING signaling in cardiac fibroblasts as a driver of late cardiac toxicity. We find delayed activation of a type I interferon response specific to cardiac fibroblasts following cardiac radiation or doxorubicin treatment, dependent on cGAS/STING signaling. Genetic or pharmacologic abrogation of this signaling eliminates the interferon response, rescues cardiac function, and improves cardiac specific mortality following DNA damaging treatments.
