Hi.... Culter maintenance can be done on chitin agar slants or plate, or Nutrient agar slants or plates. Glycerol stock is another method. Further for long time preservation one can go for lyophilization.
That depends on budget.... How much money you have in your hands to put in ?(think for each and every media components cost and necessity). Also the media composition should no hamper bacterial growth or enzyme production....So before scale up you should go for media optimization for your isolate and decide what media composition you need to use.
