The Uffizi is a huge building, so consider that it will take you around 20 minutes to enter and to start your visit on the second floor of the Gallery. If you want to visit the second floor only, you need another 1h30', so 2 hours in total. For a full visit on both floors of the Uffizi, you have to count minimum 3 hours inside. If you come to Florence for one day, all depends on the schedule of your trains. I would suggest a 2-hours visit and then an aperitivo somewhere before getting on the train.
