If you are talking about the special attack you have to complete a game with each of the ninja turtles, and on your fourth training with the Master Splinter it will unlock. You can also try to play this game with simultaneously 4 players, then one 1 completion should be enough i reckon.

He's a boss only Donnie can fight in the story mode. Like how Mikey fights Hun, Leo fights Splinter & Raph fights Casey
