A DNA microarray (also commonly known as DNA chip or biochip) is a collection of microscopic DNA spots attached to a solid surface. Scientists use DNA microarrays to measure the expression levels of large numbers of genes simultaneously or to genotype multiple regions of a genome. Each DNA spot contains picomoles (10−12 moles) of a specific DNA sequence, known as probes (or reporters or oligos). These can be a short section of a gene or other DNA element that are used to hybridize a cDNA or cRNA (also called anti-sense RNA) sample (called target) under high-stringency conditions. Probe-target hybridization is usually detected and quantified by detection of fluorophore-, silver-, or chemiluminescence-labeled targets to determine relative abundance of nucleic acid sequences in the target. Thank you so much. I have watched many videos on this topic but this is the only video that summarizes all I have learnt so far from the previous videos on this subject. The animation was spot on! :)100%
The question about how the buffer solution detached the mRNA from the column beads? I think it has to do with the breaking of the relatively weaker hydrogen bonds that exist between A-T base pair since they have fewer hydrogen bonds compared to G-C base pair. Please I hope this is right if not what is the answer?
I also have a question. Why do some microarray hybridizations use Biotin-cRNA for the labelling of the cDNA (they call it in-vitro transcription) in the sample instead of the labelling mix used in this video which would result in a labelled cDNA and not a labelled cRNA? Thank you.
