Title: Common and selective representation of pain, emotion, and cognitive control in the insula.
Session: Talk
Speaker: Mijin Kwon
The insula has been associated with numerous mental functions. However, direct evidence for functional selectivity and generalizability across insula subregions is limited. This study aims to address how insular sub-regions respond across four domains of mental processes: pain, appetitive process, aversive process, and cognitive control. First, we aggregated participant-level fMRI contrast maps from 36 studies and 540 participants. The sample was systematically constructed, with four domains, three subdomains per domain, three studies per subdomain, and 15 parti...

Re. spin locking for Glc exchange-based contrast in activation (@1:01:00), a problem I have encountered with MRS is that while the flux is high, the steady state concentration is low. (Even more so for Glc6P, which is essentially invisible in the MR spectrum even though its flux is very, very high.) So upon activation, if the Glc flux increases there is no reason to expect the [Glc] to change in concert. We need a more dynamic measure, likely involving an isotopic tracer.

Seong-Gi is one of the best fMRI contrast scientists out there. (Also one of the nicest, and probably the most humble bar none.) There is a veritable treasure trove of information here related to functional imaging contrast mechanisms. All fMRI practitioners should listen to this interview twice and take copious notes. It's a comprehensive review of what we know in late 2021.

It's becoming clear that with all the brain and consciousness theories out there, the proof will be in the pudding. By this I mean, can any particular theory be used to create a human adult level conscious machine. My bet is on the late Gerald Edelman's Extended Theory of Neuronal Group Selection. The lead group in robotics based on this theory is the Neurorobotics Lab at UC at Irvine. Dr. Edelman distinguished between primary consciousness, which came first in evolution, and that humans share with other conscious animals, and higher order consciousness, which came to only humans with the acquisition of language. A machine with primary consciousness will probably have to come first. The thing I find special about the TNGS is the Darwin series of automata created at the Neurosciences Institute by Dr. Edelman and his colleagues in the 1990's and 2000's. These machines perform in the real world, not in a restricted simulated world, and display convincing physical behavior indicative of higher psychological functions necessary for consciousness, such as perceptual categorization, memory, and learning. They are based on realistic models of the parts of the biological brain that the theory claims subserve these functions. The extended TNGS allows for the emergence of consciousness based only on further evolutionary development of the brain areas responsible for these functions, in a parsimonious way. No other research I've encountered is anywhere near as convincing. I post because on almost every video and article about the brain and consciousness that I encounter, the attitude seems to be that we still know next to nothing about how the brain and consciousness work; that there's lots of data but no unifying theory. I believe the extended TNGS is that theory. My motivation is to keep that theory in front of the public. And obviously, I consider it the route to a truly conscious machine, primary and higher-order. My advice to people who want to create a conscious machine is to seriously ground themselves in the extended TNGS and the Darwin automata first, and proceed from there, by applying to Jeff Krichmar's lab at UC Irvine, possibly. Dr. Edelman's roadmap to a conscious machine is at arxiv.org/abs/2105.10461

Summary of 1st presentation (quick overview) 2 principle fundamentales of brain organisation : - functional segregation (brain regions) - functional integration (connections between brain regions) 3 types of connectivity: - structural: anatomical - functional connectivity: correlations between signals from brain regions - effective connectivity: effect of neurons on one another Knowledge in functional segregation is much more advanced than functional integration. Map of the connectivity in the macaque brain: Felleman, Van Essen, 1991 Tools to measure brain connectivity (conectome): - Diffusion MRI (diffusion of water inside myelinated axons --> structural connectivity) - FMRI (correlations between brain regions amount of blood flows --> functional connectivity) - MEG (and EEG?) more recently Parameters of FMRI Input: heuristics to define nodes and edges Output: direction of the connection, its weight, its effect (inhibition vs excitation) Issues with MRI: head motion and respiration influence data. Processing the info: - graph theory: nodes and edges, topology
