You can Easily Find this Mushroom in your Yard (Mower's Mushroom) #mushroom #common #nature
This Species is a variety of Mushroom that almost everyone has encountered at some point. The Mower's mushroom (Panaeolus foenisecii), is such a widespread and common species, it's harder to say you haven't seen these before than it is to say you haven't. It does have some closely related relatives which some people use for recreational purposes, but this specific species is not psychoactive in any way. That being said, this is still a very good species to use to become familiar with different varieties of fungi, and to help you learn how to properly identify mushrooms.

How to find Tardigrades (While Testing a Camera Gimbal) #microscope #tardigrade #microbiology
Просмотров 775 месяцев назад
I've been wanting to try out this camera gimbal I purchased for a while. Now that the weather is starting to get a little better I decided to take a walk out in the woods in order to test it out. But because I always have to be focusing on 652 things at once I thought I would use the opportunity to look for moss to take home and try and extract tardigrades from. I, and I'm sure, you, have read ...

Pretty cool, but found 2 mistakes. First, you opened a contaminated plate right in the strongest airflow, which could contaminate your sample. Looking carefully at each plate would remedy this. Two, your sterile tek is pretty good, but it's best to spray a mushroom lightly with iso, or at least peroxide, to avoid contaminating the inside of mushroom with surface contaminants. Overall good tutorial.

I was just reading about what makes hydrogen peroxide bubble because I have an infection on my skin that causes hydrogen peroxide to foam. Everything I read says that catalase is the enzyme found in cells that starts the reaction with hydrogen peroxide. It'll foam on bacteria and fungus (yeast) and also the cells in a wound like blood cells. I found your video while going down this rabbit hole.

I was watching other videos and they use the filters of the masks both in fabric and the breathing valves of masks. Thanks for sharing your experience

If I'm doing a lot at once I'll cook them for an hour at full temperatures and pressures, then I'll let them sit in the closed pot overnight until they slowly cool down. If I'm only doing a few I'll just cook them for 25 or 30 minutes and take them out as soon as the pressure releases. They have a fairly high surface area so they heat through pretty quickly and evenly

I've been thinking about this. I like the idea of those condiment cups & I think I am going to try them. My thoughts on this is, - Pressure cook up a batch of agar, with an injection port on the jar lid. - Set up a bunch of these cups, inject agar through the lid with a sharp needle & syringe, cover hole with tape, let it sit for a week to see if any contamination grows on the agar - if the cup is clean then it's probably pretty sterile to use. - Inject through the same hole, either spores in solution or mycelium, and let it grow out. - If any contamination occurs in the cup, then it'll either have to be cleaned up, or disposed of. But if the cup is a clean grow great! - With a clean grown cup of mycelium, inject again through the same hole, as stream of sterilized water to blast the mycelium off the agar, which can then be drawn into the syringe and injected into a pressure cooked jar of whatever nutrient needed to grow it out and make more syringes. The idea is to not open the cups to the air letting contamination in, the only thing that enters through the exact same hole multiple times is a flame sterilized needle. So you could go from agar to grow jar without exposing anything to the atmosphere without a lid. (minus one tiny needle hole, that gets covered with tape. So the surface area of the agar is not exposed to the dust in the room etc. And since the agar sat for awhile without anything on it, you'll know if the container is contaminated before wasting time waiting for the mycelium to grow out. Just make up a big batch of cups, and let them sit in storage for awhile. Pull it out, ready to use, observe if there's any contamination. Inject, tape, grow out, draw off and go from there. Since there's a lid, just like the pitri dish, it can be removed in a sterile environment and then agar cut out and portions isolated as normal. One could even tape or cellophane around the edge of the cup just like is normally done with a pitri dish. I think the downside of a glass pitri dish is the fact that a needle can't get through the glass. I wonder if they make glass pitri dishes with self sealing injection port holes, or holes cut in the glass top that accept the standard injection port plugs.

I had a couple blow up on me because I filled them too much and they swelled up because they absorbed water from the high pressure steam. Try filling them slightly less than what you think they need and double seal each edge that you are sealing. I should have mentioned to fill them a little less than what you think they need because they do absorb water during the sterilizing process. That should help (:

I would imagine so. I use regular double zipper Ziploc bags for my grain. If you put a liner, I use cut up wal mart bags in your instant or pressure cooker to keep the baggies from sticking together or to the pot. The foodsaver bags are a little sturdier, should be A - ok ! Best of luck.

*"I use an Instant Pot"* *[points to a 'Gourmia', not an Instant Pot]* 😜 Jokes aside, thank you for the video! I'm foraying into mushroom growing and this is very helpful, since premade "sterile" agar plates are ridiculously expensive (and possible not sterile).

Hahaha the cheapskate version of an instant pot I guess would be more accurate. I found it at my local surplus store for 30 dollars. But thank you. I'm glad my video was able to help you. I made it for that exact reason. Pre-poured plates are way too expensive for the amount I go through. So I've always just made my own.
