No, that ball is neither a no ball nor counted as one in the over, it's a Dead ball on which the batter can be dismissed as Runout (Mankad).
What if bowler had released the ball and then the stumps were broken?
If the stumps are broken in the run-up it is a no ball even if the ball has been released
