The reason why this topic isn't more talked about is probably because there is an actual lack of evidence of the experiment actually working and creating such a breakthrough, as claimed by the researchers. The research team was anonymous, the experimental process (reportedly) wasn't documented properly, and no external research committee has reviewed and verified the research process and its results.

How can he get such substantial number of embryonic cells before returning the embryo into mother’s body? And what was the "reference" genome to check if there are potential changes of the sequence?

This actually could be fake, just to raise awareness for the controversy around human genome editing. There are some red flags: 1, There is no paper or preprint 2, Hospital doesn't know about the existence of the babies and 3, I fail to understand why you would knockout a receptor to which HIV binds if the dad is the one with the HIV. Isn't the HIV removed from the sperm when you do the IVF treatment? 4, Whole genome sequencing before and after editing is a very good idea, however it is very very hard to make 100% sure CRISPR did not aspecifically target something. Variant calling and other bam analyses would detect changes in the DNA if the depth is good enough, however if for some reason the CRISPR-cas9 creates or inserts DNA that does not align to the human genome, then it will go undetected with those analyses. You could write some scripts to check changes in the unmapped or align against other genomes like virusses and transposons that are not known transposable elements, but I doubt that they did that.

For example, what they claimed to have done here, is disabling one gene, which is easy. These experiments are highly controversial and we don't know the final results. As I know he is in prison at the present time in China due that he violated China's regulations. I think he tried to help science but did not realize that this technology may be used for different uses.
The funny thing is, that the guy KNEW about the papers on supermemory mice with the deleted CCR5 gene. The edit was made not only to make the children immune to HIV. It was partially done to make some sort of supermemory superhumans. xd
The Biggest mistake that He and his team made was that he went very public about his experiments. Given the ease of using CRISPR-cas9 technology, it's clear that there are many experiments already under way that we don't know about.
I don't understand what he did wrong, aside from practicing illegally. He made GMO children? Should be fine for people who approve of GMOs. He modified embryos? Pro-choice people dismiss potential for the future of that embryo in favor of the parent's choice. Therefore he did not "harm" the children, just as how you cannot "harm" the fetus by killing it. Also, he effectively did a procedure on humans that's barely been tried with animals; in medicine, you never try out a therapy on humans until you understand the safety, efficacy, and mechanism/rationale of the individual steps of the procedure. Also, even if gene editing was a common thing, there are far less risky procedures that would be tried before turning to editing of the embryo; for example, editing bone marrow stem cells in HIV infected patients so that you're not messing around with all the cells of a human body. Gene therapy holds enormous promise, but it has to be developed more responsibly and cautiously than this.

Dude is crazy. One of the biggest problems with CRISPR is creating perfect guide RNA so that the CRISPR system doesnt run around trying to modify all DNA sequences that are slightly similar. Off targets are the biggest problem and could cause HUGE issues. This shit doesnt seem right to me
Perhaps the rush to be first is more short-sighted arrogance than anything else. Plenty of people know how to do this, including the people that invented it at Harvard, Berkley, and a handful of other labs including one in Shanghai. The issue wasn't that it couldn't be done, but should it be done. It is a very different thing playing with a child who has no say in how they will be born, and working with an adult that can give consent. Plus there is no consideration of the additional generations this experiment affects. There is so much we don't know about our DNA, and the collateral damage this technique may cause to other genes that are dormant, but we may need in human kind's future evolution.
So with the argument about "consent", should we stop circumcising babies? What about implementing religion or giving them a name they didn't choose? So, let's ignore that issue. We can never predict the effects of our actions. We didn't foresee the climate impact of industrialization, but you wouldn't be typing on a tiny computer that fits in your pocket if we didn't pursue it. So let's ignore "considering the future effects" cause we can't predict that. And technology naturally affects our species' evolution. Discovering fire and using it to cook your food, is the same technological advancement as using CRISPR. So let's ignore that argument too.

However, among scientists in the field we are concerned about long-term effects of gene-editing. We aren't sure if there are any unintended consequences of editing certain genes in or out or whether this technology is as precise as we wish it to be. Combining that with how much we don't understand about the whole system we're manipulating and it could be dangerous in the long run.

The only bad thing is, kids don't have a say on the matter. It's like feeding your kids/babies vegetables. You know the benefits of it, but they dont. What do you say to that? And also, it's hard to predict the effects of our action. Did you know we don't know how analgesics work? Yet Tylenol is over the counter. Look it up! I can remember back to around the mid 90's when they first cloned the Sheep: "Dolly" Immediately after they were successful, two Italian doctors sought permission to clone Human Beings. They were refused. But who's to say that they didn't sneak off some place and do it anyway. I mean, when the W.H.O ordered all doctors and pathologist not to perform Post Mortems / Autopsies on C.O.V.I.D-19 deaths. Italian doctors and German Doctors didn't listen to the orders, and then they came out and spoke openly of their findings.

In this very specific case (of "Lulu and Nana"), there is a reliable method to ensure whether their DNA was harmed inadvertently in any areas other than that intended (the CCR5 "HIV-infection" gene). It is not clear whether Dr. He followed this method or not - he was unclear in his presentation. It could be proven safe in this specific case IF he did ALL of the following: 1) he sequenced the zygote's ("Lulu / Nana"'s) entire DNA before introducing the gene-splicing modification (the "Original" DNA), 2) After introducing the gene-splicing modification, he sequenced the entire DNA (the "Modified" DNA). 3) Do a full comparative analysis of the differences between the "Original" DNA and the "Modified" DNA -- effectively subtract/compare the two full sequences of DNA looking for any/all variations. 4) Ensure that the ONLY variations in the "Modified" DNA are to the CCR5 region. If his lab was able to demonstrate step 4, with high statistical significance and precision, then we can be sure the girls will be safe from unintended genetic harm. We then will have to wait and see if they will be immune to HIV-1 infection. (though, they may not be immune to other forms of HIV). This explanation provided by Dr. He's lab was very important to understanding what was done and why. However, this is only a verbal statement. We need his statements to be verified through independent means: a) is it true that the genetic father of "Lulu and Nana" is HIV-positive b) does the genetic father have the HIV-1 variant? c) Can other labs also sequence "Lulu and Nana"'s DNA to confirm the sequencing done by Dr. He's lab was correctly done and is reproducible/confirmed Though this is a controversial area, scientists can implement proper stringent controls to ensure such work, on individual cases, are done with sufficient controls to ensure safety (societal and individual).
To be really sure about this method, it needs statistics of at least a hundred of children. But society sucks, and doing so illegally would be impossible, since no one would see any profit.

I understand your sentiment but it has been discovered that He did not work correctly with the Crispr gene editing. Not only are Lulu and Nana not protected against HIV but *are* careless science experiments with unknown outcomes for themselves and future generations they may birth.
Unfortunately it didn't work and might even caused them to be more susceptible to other illness. It was super complicated but if you read the newer info about them, it's scary and sad.
Are the twins now immune to HIV? If so, can they pass those genes onto their children? If so, doesn't that mean eventually humanity will be immune to HIV if their genes manage to spread into future generations? If they each had 2 children and the average birth rate of their subsequent offspring was 2, then we would have over 2 billion humans born with HIV immunity after 30 generations.
Yes, this was so-called germline editing, so in principle the trait of immunity to HIV as we know it will be passed on to any offspring. Other than that, remember that the human species first has to manage to survive before you can get to your 30 generations. It's also possible that the HIV virus no longer exists by then.

The problems being: one, there is no breakthrough and two, it’s not ethical. The technique has been used for years and close to being patented, just look it up. It´s not that easy, because one thing He Jiankuil did´nt mention is that CRISPR did not work as planed and the babies suffered genetic mosaicism for CCR5 (the gene he talks about in the video).
I am quite indecisive whether this could be the dawn of a utopian (i.e. fatal trait removal, ethical enhancement) or a dystopian (e.g. Auxley's Brave New World's alpha/beta/.../epsilon-castes) society for humankind. Possibly somewhere between the two extremes, but it's both inspiring and frightening.
The CCR5 gene he edited (or rather disabled) is correlated with memory and intelligence. It is absent in the Chinese population, but prevalent in northern Europe.
A world first for Prof He Jiankui. Many world firsts have come and gone. Some have propelled humanity to greatness, others not so. You claim good intentions but we'll let history be the judge. Perhaps some day down the road, this will all be behind us and you'll have the Nobel in your hand. I hope and pray that the Lulu and Nana will be there when that day comes.

If you support this guy, then you clearly did not look at the full extent of the risks and implications of his experiment. 1) The experiment was not approved, first of all. 2) He did not inform the parents of the risks. 3) He created frameshift null mutations because the CCR5 alleles were not created in the twin girls. As a result of this, one of them is mosaic (this means that it was genetically different from the twins, which can cause disease in the future) and the girls may still susceptible to HIV. 4) The scientific basis for the study is weak: it is very rare for fathers to pass HIV down to daughters. It's difficult to say, but there might be other mutations and genes altered that might impact the girls in the future. Please be aware of the conflict in ethics and safety of this study.

The reason why this topic isn't more talked about is probably because there is an actual lack of evidence of the experiment actually working and creating such a breakthrough, as claimed by the researchers. The research team was anonymous, the experimental process (reportedly) wasn't documented properly, and no external research committee has reviewed and verified the research process and its results.

As far as I know (sorry, I am not a specialist in this field so I might have misunderstood) the whole genome sequencing to find one nucleotide change requires substantial numbers of cells to normalize the mis-reading of the sequence. How can he get such substantial number of embryonic cells before returning the embryo into mother’s body?
And what was the "reference" genome to check if there are potential changes of the sequence?
The cell multiplied before it was put back into the womb, as embryos often do.

This actually could be fake, just to raise awareness for the controversy around human genome editing. There are some red flags: 1, There is no paper or preprint 2, Hospital doesn't know about the existence of the babies and 3, I fail to understand why you would knockout a receptor to which HIV binds if the dad is the one with the HIV. Isn't the HIV removed from the sperm when you do the IVF treatment? 4, Whole genome sequencing before and after editing is a very good idea, however it is very very hard to make 100% sure CRISPR did not aspecifically target something. Variant calling and other bam analyses would detect changes in the DNA if the depth is good enough, however if for some reason the CRISPR-cas9 creates or inserts DNA that does not align to the human genome, then it will go undetected with those analyses. You could write some scripts to check changes in the unmapped or align against other genomes like virusses and transposons that are not known transposable elements, but I doubt that they did that.

They think homosexuality has more to do with epigenetics than with genetics, which makes it a lot harder to modify. But still, changing the whole mechanism of something is really hard. For example, what they claimed to have done here, is disabling one gene, which is easy. But when you want to change something like the way an eye functions or the when someone gets angry, there are so many processes involved. You would first need to map tens of genes involved in the cascade and then know how they work and know how you can change them for the better. It is soo many information and right now we do not even know what most genes exactly do, let alone in which cascade they belong and how they exactly function. Now because homosexuality is even more complicated than that, I do not think that we will live to see that ppl can engineer sexuality.

Also, he effectively did a procedure on humans that's barely been tried with animals; in medicine, you never try out a therapy on humans until you understand the safety, efficacy, and mechanism/rationale of the individual steps of the procedure. Also, even if gene editing was a common thing, there are far less risky procedures that would be tried before turning to editing of the embryo; for example, editing bone marrow stem cells in HIV infected patients so that you're not messing around with all the cells of a human body. Gene therapy holds enormous promise, but it has to be developed more responsibly and cautiously than this.

I'm against the fact that he was jailed in China. Parents had already given their consent so it's wrong what was done to this doctor. I hope it was a deletion mutation. So, no other genes were alerted. Well, those girls were never followed up for their progress, so it's unknown what happened to their health.
Dude is crazy. One of the biggest problems with CRISPR is creating perfect guide RNA so that the CRISPR system doesnt run around trying to modify all DNA sequences that are slightly similar. Off targets are the biggest problem and could cause HUGE issues. This shit doesnt seem right to me
Perhaps the rush to be first is more short-sighted arrogance than anything else. Plenty of people know how to do this, including the people that invented it at Harvard, Berkley, and a handful of other labs including one in Shanghai. The issue wasn't that it couldn't be done, but should it be done. It is a very different thing playing with a child who has no say in how they will be born, and working with an adult that can give consent. Plus there is no consideration of the additional generations this experiment affects. There is so much we don't know about our DNA, and the collateral damage this technique may cause to other genes that are dormant, but we may need in human kind's future evolution.
So even in this case, where these children were born without HIV, you wouldn't even consider any benefits from this?
So with the argument about "consent", should we stop circumcising babies? What about implementing religion or giving them a name they didn't choose? So, let's ignore that issue. Discovering fire and using it to cook your food, is the same technological advancement as using CRISPR. So let's ignore that argument too.

However, among scientists in the field we are concerned about long-term effects of gene-editing. We aren't sure if there are any unintended consequences of editing certain genes in or out or whether this technology is as precise as we wish it to be. Combining that with how much we don't understand about the whole system we're manipulating and it could be dangerous in the long run.

Thanks for your reply @@boukmann1791! I looked in multiple articles online and searched in published papers and apparently circumcision prevents UTI on babies. Circumcision is a good thing. The only bad thing is, kids don't have a say on the matter. It's like feeding your kids/babies vegetables. You know the benefits of it, but they dont. What do you say to that?

I can remember back to around the mid 90's when they first cloned the Sheep: "Dolly" Immediately after they were successful, two Italian doctors sought permission to clone Human Beings. They were refused. But who's to say that they didn't sneak off some place and do it anyway. I mean, when the W.H.O ordered all doctors and pathologist not to perform Post Mortems / Autopsies on C.O.V.I.D-19 deaths. Italian doctors and German Doctors didn't listen to the orders, and then they came out and spoke openly of their findings.

In this very specific case (of "Lulu and Nana"), there is a reliable method to ensure whether their DNA was harmed inadvertently in any areas other than that intended (the CCR5 "HIV-infection" gene). It is not clear whether Dr. He followed this method or not - he was unclear in his presentation.
It could be proven safe in this specific case IF he did ALL of the following:
1) he sequenced the zygote's ("Lulu / Nana"'s) entire DNA before introducing the gene-splicing modification (the "Original" DNA),
2) After introducing the gene-splicing modification, he sequenced the entire DNA (the "Modified" DNA).
3) Do a full comparative analysis of the differences between the "Original" DNA and the "Modified" DNA -- effectively subtract/compare the two full sequences of DNA looking for any/all variations.
4) Ensure that the ONLY variations in the "Modified" DNA are to the CCR5 region.
If his lab was able to demonstrate step 4, with high statistical significance and precision, then we can be sure the girls will be safe from unintended genetic harm. We then will have to wait and see if they will be immune to HIV-1 infection. (though, they may not be immune to other forms of HIV).
This explanation provided by Dr. He's lab was very important to understanding what was done and why. However, this is only a verbal statement. We need his statements to be verified through independent means:
a) is it true that the genetic father of "Lulu and Nana" is HIV-positive
b) does the genetic father have the HIV-1 variant?
c) Can other labs also sequence "Lulu and Nana"'s DNA to confirm the sequencing done by Dr. He's lab was correctly done and is reproducible/confirmed
Though this is a controversial area, scientists can implement proper stringent controls to ensure such work, on individual cases, are done with sufficient controls to ensure safety (societal and individual).

To be really sure about this method, it needs statistics of at least a hundred of children. But society sucks, and doing so illegally would be impossible, since no one would see any profit.

To me, this man is a hero. If you want to achieve big scientific breakthroughs, you need to be bold. Sanctimonious bioethicists love to express "outrage and shock" at this type of work, but what have they done? The problems being: one, there is no breakthrough and two, it’s not ethical. The technique has been used for years and close to being patented, just look it up. It´s not that easy, because one thing He Jiankuil did´nt mention is that CRISPR did not work as planed and the babies suffered genetic mosaicism for CCR5 (the gene he talks about in the video).

I am quite indecisive whether this could be the dawn of a utopian (i.e. fatal trait removal, ethical enhancement) or a dystopian (e.g. Auxley's Brave New World's alpha/beta/.../epsilon-castes) society for humankind. The CCR5 gene he edited (or rather disabled) is correlated with memory and intelligence. It is absent in the Chinese population, but prevalent in northern Europe.
A world first for Prof He Jiankui. Many world firsts have come and gone. Some have propelled humanity to greatness, others not so. You claim good intentions but we'll let history be the judge. Perhaps some day down the road, this will all be behind us and you'll have the Nobel in your hand. I hope and pray that the Lulu and Nana will be there when that day comes.

If you support this guy, then you clearly did not look at the full extent of the risks and implications of his experiment.
1) The experiment was not approved, first of all.
2) He did not inform the parents of the risks.
3) He created frameshift null mutations because the CCR5 alleles were not created in the twin girls. As a result of this, one of them is mosaic (this means that it was genetically different from the twins, which can cause disease in the future) and the girls may still susceptible to HIV.
4) The scientific basis for the study is weak: it is very rare for fathers to pass HIV down to daughters.
It's difficult to say, but there might be other mutations and genes altered that might impact the girls in the future. Please be aware of the conflict in ethics and safety of this study.
