    When I first started watching Hamilton's Pharmacopeia, I didn't know who he was and I thought that him reciting chemical compounds and processes during the montages was just a gimmick. I swiftly realized the man is a brilliant chemist. He literally synthesized a compound because he wanted to preserve a population of desert toads so people could get high sustainably without harming them. Wild.
    Edit: To everyone telling me how easy this synthesis is, I bet your Big Dick chemistry degree from Shitballs University was a breeze, it's incredible how many "chemists" come out of the woodwork when you compliment one on RUclips

      As someone who has taken organic chemistry on the college level I can't discern whether he's a brilliant chemist. He could 100% have a good understanding of college level organic chemistry and give this kind of presentation. It doesn't make him a brilliant chemist. I'm not saying he's not, however. He could be. It's just that it's not obvious to me as someone with a college level understanding of orgo could present this way.

      @@HexFent Ay bro, ScienceGuy actually has a really good point. I myself am doing a bachelors in Chem. and Hamilton sure does sound like he has some knowledge about chemistry but not to the level of a genius. You could probably give anybody an Alexander Shulgin book or even an intro chem textbook and if you regurgitate that information to a regular person with little chem knowledge, they'll think you're a genius too but when you have a background in chemistry, the stuff he speaks about is actually pretty basic.
      I think most people can understand the topics he brings up by spending a few hours reading up articles online or a textbook, nothing ground breaking enough to be considered a genius, but I will admit that he has more knowledge about it than the general populace.

      @@NolieRavioli My guy, I was referring to how OP was saying he's a brilliant chemist. Neither me or scienceguy was saying that Hamilton is not a smart guy, just maybe not a brilliant CHEMIST, as we can't tell simply from the chemistry he uses in his videos and documentaries. So in this case, we really ARE talking about his knowledge in the "classical sense" since OP literally calls him a brilliant chemist.
      He is definitely a smart guy and I have been watching his stuff for years. Pay attention more, not everybody is out there to talk shit or to criticize, we aren't calling him out. I love his stuff and his knowledge/ability in transformative content far out stretches mine, I doubt I can make a documentary half as good as his, but calling him a genius chemist when he shows a reaction scheme (as an example) that he didn't write out himself even is a bit far fetched.

      @@laurean5998 you don't need large amounts of methanol unless you want to make 10s or 100s of grams of the stuff. Considering the dose is around 5mg you wouldn't need much.
      But why do all that when you can just extract n,n-dmt, it's much simpler and a way better drug

      I wish they were readily available in my place.
      Microdosing was my next plan of care for my husband. He is 59 & has so many mental health issues plus probable CTE & a TBI that left him in a coma 8 days. It's too late now I had to get a TPO as he's 6'6 300+ pound homicidal maniac.

      I loved the theory, but screwed up the practical. I could follow recipes to cooking classes, but not the steps in a lab. Going to have to do A LOT of research and relearning things before making DMT & changa this year.

What he says is that he doesn't want to/can't put a step by step guide out there so people can't just follow how to do it like a recipe in a cook book. Good luck with your endeavour tho.

    This chemical synthesis is the so-called reductive amination by the Eschweiler-Clarke reaction using 37% formaldehyde and conc. formic acid. 86%, the reaction has been modified by replacing formic acid as a hydride source with sodium cyanoborohydride (NaCNBH3). A solution of 5-methoxytryptophan in anhydrous or dry methanol (100% free of water) is added to the reaction balloon with an ice bath, then the 37% formaldehyde solution is placed in an addition funnel and in another addition funnel a solution of sodium cyanoborohydride dissolved in methanol or dry ether or tetrahydruphorane (THF), both solutions are added drop by drop with constant stirring determining that the temperature does not increase, it can be helped with a reflux condenser. Once the reaction is finished, the crude oil is separated from the 5-methoxytryptamine and a fractional distillation is carried out, the distilled psychotropic is re-purified by a simple distillation, it is crystallized by dissolving the psychotropic in methanol or toluene, the crystals are cooled and filtered. It can be obtained from melatonin by deacetylation carrying out a reaction under reflux with hydrochloric acid and neutralizing with sodium hydroxide until alkaline pH (7-9), the precipitate is filtered and the reductive amination is applied.

      @@samuelsbald3038 both are good reducing agents, what happens is that sodium borohydride is much more reactive than sodium cyanoborohydride and is used more in reductive amination like the other, the difference is that sodium borohydride being stronger is capable of directly reducing formaldehyde to alcohol, while the other is not because it is less reactive, so that this does not happen, it is better to react the amino group with formaldehyde for 1 hour to form the imine, and then reduce it, but both are very good, you just have to keep that in mind.

      Only a small mistake, but "5-methoxyTRYPTOPHAN" (5-meo-TP) is not the compound you would use for this synthesis, instead you would start with "5-methoxyTRYPTAMINE". 5-methoxytryptophan in this reaction would yield '5-methoxy n,n-dimethyltryptophan'. It is very very likely that this compound produces psychedelic effects, but has not been studied enough to be sure of that.
      If starting from 5-meo-TP a decarboxylation would have to be carried out first in order to yield 5-meo-T. You could use Lithium Aluminum Hydride (LiAlH3) as a reducing agent for this reaction. Then from here, you would have to purify the 5-meo-T via recrystalisation. With your 5-meo-T you could do a reductive amination by the Eschweiler Clarke reaction to yield 5-meo-dmt.

    Hi Hamilton, I just finished my last year of high school, your documentaries and vice articles (especially the sea sponge one) have been a huge influence on my interest in chemistry. Although Covid cancelled many of my in class practical experiments, I have had a lot of fun creating various esters and interesting smelling compounds at home.     Macro vs Micro had success in doing a few Macro first . Allowing the mushrooms to blast you into self awareness , showing you the shadows and trauma that have been hiding under the rug . Micro dosing has helped me stay on the pulse of my shadow work , helping me remember my best self .

      @@czlenson888 1) The plant you're referring to is called mimosa hostilis (not misosa), it mostly contains N,N-dimethyltryptamine (DMT), which can be extracted using an acid base extraction, (which is essentially what you're describing).

      He uses a lot of fancy chemistry terms. But all you would really need is the starting chemicals, about $100 worth of glassware, and maybe a hot plate and the patience to start all over if you don't get it right. Google some of the chemistry terms then it's basically like baking a cake.

    Thought this may interest anyone it's a story of my dmt cartridge experience :). So about 6 hits into the cartridge eyes closed I feel as if my mind now has became almost a painting that's been turned on but not fully activated. Space itself was flashing in my mind like if I was blinking inside my mind but every time a new brief image of galaxies stars etc appeared. And I could feel it with 100% of my senses those I didint quite even understand lol. As the body high increased with my hits I began to notice a feeling as if a thin "Soul" was being flashed out of my body and it was telling me this for certain when I came into a realm of beautiful darkness highlighted by a dark "wall" of deep deep tunnels with rims resembling black holes with rainbow chromish edges. I then felt the intensity of a specific tunnel and could feel its depth before I even went inside but when I did I was transported to a world where I looked at a bigger version of myself cut in half with living mechanical gears perfectly in place like a puzzle in my brain and other organs. And then as I became one with this creation in another flash I was flying through a rainbow tornado on an earth type planet but the clouds blocked my vision of most of that planet pretty sure 😭.

      The extraction methods that previous commenters mention is for N,N-DMT, which is slightly different than the 5-MeO-DMT we see Hamilton synthesizing here. Both are some of (if not the) strongest psychedelic substances we know of at this time, but from what I've read, 5-MeO is generally regarded as more intense than N,N (I've only done N,N, so I can't attest to this, personally). The extraction process for N,N-DMT is fairly straightforward, but as with all things dealing with chemicals, should be treated with respect and precaution.

    I just finished my second semester of organic chemistry at GVSU. I happened upon your show a few months in to my 1st semester last year and watched every episode, it's rewarding to be able to understand (for the most part) the many mechanisms discussed in the reactions in your show. Additionally, it's amazing how ultimately all the complex and enigmatic psychoactive effects from these compounds boil down to the infinitesimal transfer of electrons.
    I think this show is giving many young people the spark, the inspiration they need to fuel their passion for learning science, and it is bringing light to the wondrous field of chemistry.

      you can deacetylate it with naoh solution , heat to 105c in isobutanol for a few hours reduce the solvent down 50% , add 10% acetone and freeze it, youll get a bunch of good crystals , you can repeat it to scavenge the rest of the mexamine from solution on a second crystallization.

    I was involved with a vivacious group of people in the early 2000s that ran a true clandestine LSD lab I was dating a woman who's brother was a organic chemist at the University of Washington at the time we all lived in the "U" district and it was the most compelling time of my life. Long story short I agreed to be the fall guy when the feds were tipped off they ransacked the house found only a few legal precursor chems a 40 bag of weed. What they didn't even realize is that on the wall there was a 48x30 self portrait of van gogh made of pure blotter it was mostly inert due to light, oxygen exposure but goddamn what a wild ride I wish there were time machines!       @@ME-ol9gk I believe there are comprehensive resources on Reddit available to guide you. It's relatively safe to use the darknet when you follow directions carefully. I personally had spent thousands in the past with out issue.

      @@chigbungus5257 Psychedelics like psilocybin, DMT, LSD etc. tend to affect your sense of self; your ego, the 'voice in your head.' Altering or obliterating that voice, even temporarily, can be risky. Depending on your starting point (and few really are so self-aware that they know themselves that deeply), you cannot be sure if the change will be one you want or can live with easily. Those that find it positive and magical sing it's praises, but less often heard are those that do not come back, and instead trigger a latent psychosis or other negative effect.

      Nice video but the mecanism he gives is kinda wrong, he's suggesting a cationic protonated alcohol leaving group during his reductive amination. You're never gonna form that in the presense of sodium borohydride (H-). Sorry, annoying O-chem PhD here :D

    You're amazing. Keep doing your thing. For some reason when I saw the cuts to you eating a banana after talking about the synthesis, I thought about how much perspective matters. You're doing amazing things with chemistry, and then you cut to eating a simple banana. When I personally thought about it, I thought that both scenes are incredible in their own right. Obviously you synthesizing 5-meo-dmt, but also just how amazing it is that we're here and have bananas to eat. Depending on your perspective at the moment, both could seem uninteresting and dull. I was wondering if you are willing to continue your research on the synthesis of tryptamine derivatives. The 5-MeO-tryptamine as a starting material is not accessible to a lot of chemists and industries around the world. How about the complete synthesis of DMT or other useful tryptamines from an indole or tryptophan as a starting material? The literature is very old and with a lot of mistakes.

    For those who want to do further research on this reaction, this is the Eschweiler-Clarke reaction, which is used for the selective n-methylation of primary and secondary amines. But the reaction traditionally uses highly concentrated formic acid instead of sodium borohydride.
