@Microbiology Tube hi i have a few questions. first on which gelose did you carry out the confrontation tests (antibiograme actinomycetes and target bacteria)? how do we name this technique and how it was done please i saw the video but i did not understand the method thank you .
It seems to be their is high chance of contamination try to do it in the LAF so we get better result . And their is mistake in handling the pipette and while taking the rod from the acetone pls wait for few seconds and then expose into the flame .
There are few things you actually need gloves for. And it also depends a lot on what you’re trying to protect. Yourself...or the sample. Wearing gloves when they’re actually not needed just makes you careless and sloppy. (That said at that point they’re working s1 with soil samples. They’re working sterile (next to the flame - where wearing gloves actually endangers you; melting plastic and all that)) so that’s a clear no to your question
we used ISP 4 medium. We tried with starch casein agar, however, ISP 4 medium gave better result. ISP 4 medium can be prepared in laboratory. You can research in google for ingredients of ISP4 medium. Best of luck.

How can we do the growth curve studies as actinomycetes grow like moulds in liquid broth....how can we take spectroscopy values??

Ur not maintaining aseptic condition snd then how could you get pure colonies it might be contaminated

No i donot use any antibiotics in SCA. In my experience, ISP 4 medium is better for isolation of actinomycetes.
