Haemoglobin is the major component of blood which carries oxygen to the tissue for various function. when that goes down below the normal level one feels lethargy and fatigue. it's always better to estimate the haemoglobin level in blood.
After watching this video you will be able to understand- what is haemoglobin test, haemoglobin experiment, haemoglobin in Hindi, Sahli's hemoglobinometer, how to check haemoglobin in blood, haemoglobin estimation test, find your haemoglobin count, haemoglobin level in man, haemoglobin level in the woman,
Haemoglobin can be estimated by two methods-
Sahli's Haemoglobinometer and haemoglobin colour scale.
Step by steps procedure to estimate haemoglobin in blood by Sahli's method.
1- Take out the Haemoglobinometer and arrange its component in a stable place.
    2.Prepare 0.1N HCl
3.Fill the 0.1 N into Sahli Haemometer tube up to 2% mark
    4. Now place the pipette in Sahli Haemometer at a given place (in between two windows)
    5. Now sterilize the fingertip and prick it with the help of sterilized lancet and fill the blood into Sahli Haemometer pipette up to 20 microliter marks.
6.Now transfer that blood sample into Sahli Haemometer tube in which we have already taken 0.1 N HCl.
7.Wait for about 10 minutes to let the reaction occur.
8.Now slowly-slowly dropwise add distilled water to its side and shake well with the help of given glass rod.
9.Keep adding distilled water till the colour of blood match the side window
    10. Observe and note down the reading, that’s the percentage of haemoglobin

Just a note: The stirrer should be lifted when comparing the colors, but not completely taken out of the tube. It's so as to avoid any potential loss of fluid which would lead to a lower Hb estimate.

The finger should not be squeezed to take out the blood, otherwise tissue fluid might come out, thus giving the lower estimation of haemoglobin

    • @Joeythegoats
      @Joeythegoats 5 лет назад +58

Yeah but most of the cases we have to squeeze the finger to get enough blood at 1 time otherwise bubble will produce which will largely effect it and the blood in capillary tube may clot in no time.
When we performed in lab most of students had to squeeze it or have to prick at different place on finger to get enough blood. With a tiny prick

But u have not mentioned after placing blood in N/10 HCl in sahlis tube wait for 10 minutes
    And also
In no case we don't have to take the stirrer out, add distilled water but don't take the stirrer out as it will contaminate the solution.

Some laboratories multiply the meter reading with 8 and 9 to get the result in decimals
    i,e:-
    Meter reading.....13
    13×8= 10.4 gms%
    I still have a doubt
