thanks for the video. Normally do you make this process without facemask? I am a MD and I am worry about the posibility of contamination. Thanks again

Hi Alami, For most applications, we can purify exosomes in the lab without a flow cabinet or facemask. If you were using them in downstream applications that require sterility (e.g. in cell culture) then you would need to use a flow cabinet. We wouldn't usually use a face mask unless the samples are high risk - we don't handle those types of samples in our labs.

Great. my question is dye is color full so you collected it in 6-7 fraction. While exosomes samples generally colorless, then how we can take them on fraction tube.

Thanks for the answer. I have one more question.
    when Precipitation(B. 5step, page 7, 16000xg 1h), did you use ultracentrifuge?
we haven't ultracentrifuge. could I divide the sample into 1.5ml tubes into several and used rotate microcentrifuge?
In fact, after experimenting like this, I couldn't see the pallet. What should I do?

Hi Lily, You don't need to use an ultracentrifuge. This is one of the benefits of this technology. Please send your message to tech@cellgs.com and we can provide further advice over email.

    hi~ I have a few questions.
    1. What is the minimum volume of human CSF we need to mention if we want to get exosomes?
    2. How long does it take to receive one fraction?
How do you confirm that all solutions have been received without dye solution and move on to 2 fractions?

      Hi Lily.
      1. CSF has relatively low amounts of exosomes - much less than blood. So you'll need more. It depends on your downstream applications. There's a protocol specifically for CSF on page 7 of the protocol cellgs.e2ecdn.co.uk/Downloads/EX01_Exo-spin_User_Guide.pdf
      2. The whole Exo-spin protocol takes a few hours, but the hands-on time is much shorter. Each fraction will take less than a minute to elute from the column.

What is the minimum volume of human plasma/serum we need to mention if we want to get exosomes?

Hi Minjin. Serum/plasma are very rich in exosomes. You will be able to recover sufficient material for downstream applications from 100 ul
