Normally You can keep it always open, but if ink is under maximum level in the tank and cartridge still ink leaks from cartridge nozzles after a while, than you can keep the lock closed and just open it while printing.

Ink tank mai two chamber minimum hote hi hain. 1.Ink Chamber 2.Air vents/chamber tank ki design/functions different hoti hai to uspe depend karta hai ki kese use karna hai.
NORMALLY the lock should be open always. only need to close while installation procedure or transportation or if you want to remove cartridges from printer.
