So when you add the gellan gum to the deli containers, you don't have to mix it before autoclaving? Does it just mix evenly when boiling during the sterilizing process?
It would be best to mix it before and even after autoclaving (by swirling the media before purring it into culture vessels). This will ensure that the gelling agent is homogenously mixed into the media.
I'm all in. Been watching every video since it all began. Great to see Shay and fungi. I've never seen this cultivated. Best wishes to you all from Perth Australia
i also recently came to plant culture from fungi culture, and i as well never bothered with the PH for the fungi. But once i started learning more about PH i got curious about applying similar growing methods with fungi.. so far ive made too few experiments with too broad results,, so still hard to say about the need for controlled PH with fungi :)
Thanks for sharing.. Just wondering, can we keep the un-autoclaved liquid MS media (without agar) for more than 24 hours? When we are dealing with some different types of media, it will be easier if we have the MS liquid stocks ready beforehand
Adding more agar makes the media hard and does not allow the diffusion of nutrients into the tissues. You need to add just the right amount of agar and mix it well.
Thank you, so basically start over if media dosen't come out? I added 1/4 tsp to 83ml cups. I made 12 instead of 10 as that is how many I can put in autoclave. Thanks for your help
@@PlantCellTechnology yes i did Sir. but i really need a specific video on aquatic plant as like as Montecarlo/ Ludwigia glandulosa/Anubias/rotala/Hygrophila🙏
Please please please do a video on embryo extraction/rescue for us. Would happily pay for private video as the litterature is very broad on this topic. Pretty please 🙏 😅
LOVE THE NEW SERIES. I WANT TO SEE A MUSA TC video!

Is your class live? I’m out of town this weekend but would still love to take it.
Yes, we have an online class coming up that you can join: plantcelltechnology.com/pages/master-classes

Generally, there are chances of contamination due to sugar, so we don't recommend that.
If it dosen't gell completely can you add more agar and re run in autoclave?

@@davesharp8994 You need 8 grams of agar for 1 liter of the media, to be exact.
Sir can you please do TC video on aquatic plant please 🙏
We do have some videos on our channel. Did you check them out?

What if were using dwk media
You need to be sure that the plant you want to culture has that requirement. It is better to follow a protocol.

@@MDHTelevizion You can try MS media supplemented with suitable hormones. That has worked in our class so far. Didn't try DKW.
Do yo have protocols in pdf or video for Aroids??? Such a great job, crongrats!!!
@@ozmalo472mtz7 Currently, we don't have any, but you can find a lot of literature online.
