Sir, I am isolating plant dna from leaf sample and getting smear in it... Even after RNase treatment and purification of the DNA. Could you please provide suggestions regarding the removal the smear... Thank you in advance 🙏
Hi, the palette is usually white. However, sometimes the palette retains some colour from the original sample material. This usually happens when a large amount of fresh material is used or when the cells contain a lot of pigment. According to the manufacturer, Qiagen, 3-30ug of DNA is typical. However, my experience is that it is often greater. The yield for the technique varies a lot and may depend on the genome size of the plant you are working with. If you need a higher yield, you can increase the concentration of the DNA in the final elution by decreasing the volume of elution buffer you use. (I'm sorry for the very late reply! @@SaumyaSharma123 Yes, we routinely use the Plant Minikit for fungal DNA extraction. A few standard questions: Have you confirmed that the bee abdomen you are extracting from do contain the spores? Are you able to pool your samples during extraction to increase chances of obtaining DNA? I assume that you have seen the paper Ingemar et al 2013 Standard methods for Nosema research (doi.org/10.3896/IBRA.1.52.1.14)? The paper shows a few additional steps in the Nosema DNA extraction protocol before using the Plant Minikit.

Yes, it can. However, an animal tissue extraction method would also be effective for fungi.
