Passaging iPSCs with gentle cell dissociation reagent (GCDR)

  • In this video, we are going to go over the protocol to passage induced pluripotent stem cells (iPSCs) using gentle cell dissociation reagent (GCDR). To seed the cells, use a Matrigel-coated dish, prepared using the method we discussed in the coating video. Don’t forget to aspirate the matrigel solution from the dish right before seeding the cells.
    List of Materials:
    - Phase-contrast microscope
    - Permanent marker
    - Matrigel-coated dishes
    - Gentle cell dissociation reagent (GCDR)
    - DMEM/F12 with Antibiotic Antimycotic (Warmed to 37 ºC)
    - iPSC Media (mTSER or E8) at room temperature
    - 5 mL and 10 mL plastic and glass serological pipettes
    - Sterile pipette tips
    - Cell scraper
    - 15 mL conical centrifuge tube
