Passaging iPSCs with gentle cell dissociation reagent (GCDR)

In this video, we are going to go over the protocol to passage induced pluripotent stem cells (iPSCs) using gentle cell dissociation reagent (GCDR). To seed the cells, use a Matrigel-coated dish, prepared using the method we discussed in the coating video. Don’t forget to aspirate the matrigel solution from the dish right before seeding the cells.
List of Materials:
- Phase-contrast microscope
- Permanent marker
- Matrigel-coated dishes
- Gentle cell dissociation reagent (GCDR)
- DMEM/F12 with Antibiotic Antimycotic (Warmed to 37 ºC)
- iPSC Media (mTSER or E8) at room temperature
- 5 mL and 10 mL plastic and glass serological pipettes
- Sterile pipette tips
- Cell scraper
- 15 mL conical centrifuge tube
