    Are you using the same tip for the cck8 treatment? I dont see you changing tips in between well treatments and thats a no no especially since youre dipping back into your stock solution.

      You're right. Each group needs to change the tip, and we actually do. It's just to shorten the video duration, so we didn't show this detail in the video.

    WHAT IS MEANT BY NC GROUP

      It depends on your grouping settings, and the following information is for your reference:
      NC in CCK-8 experiments usually refers to normal cells that have not been treated, although if you are dealing with something like lipid transfection, NC usually refers to the group that has been transfected with NC fragments.

      You can use any drawing software that has the function of fitting curves.If the effect of the drug or intervention is to inhibit cell viability, and we usually want to get the IC50, then our abscissa will be the specific drug concentration, and the ordinate will be 100% minus the growth rate in the video.
      For example, we can assume that the concentrations (mM) of NC and Test1~4 groups are 0, 1000, 100, 10, 1 respectively, then the data information we get is shown in the figure:(I'm sorry that we can't send picture in the reply.       Then they are entered into the software, the pictures and formulas are obtained, and the IC50 can be calculated using the software, the following is the diagram with “CurveExpert1.4” as an example.
      Right-click on the graph to select “Analysis”, and the IC50 can be calculated.
