In my understanding yes, you just pick a spot but it has to be on the horizon. even if it's outside the borders of your canvas it should still be on the horizon plane.
There can be many vanishing points in an image, depending on where the images was taken, in which direction, with which kind of lens and station point (most important). If you have a reference image and want to find out where all the vanishing points are, I recommend you to use the pecil as a measuring stick .
