No, he wasn't. By 2016 he already had stage 3 colon cancer, no one is saved from that, he was already sure that the only thing he could do was to extend his time a little. By this time he was getting weaker, and he looked at her in the eyes seriously when she said "I'm dead" without making any joke or laugh about it (besides, there were already plans to revive his character so there is no point in answering about future events of Black Panther). No, he already knew and only a naive wouldn't realize that.

God I want to be brave like him, when my time come. He was a very beautiful and great man, there is none other like our King RIP brother.
I admire that in a world of over-sharing, he kept his health private and only between a trusted few in his circle. I've been seeing that a lot lately with people-- keeping their private lives private. It is healthy to keep others' voices out of your life sometimes, and to carefully select only a few who you trust with your private life.
