    Can you please elaborate @12:30 on protein synthesis blockage by Tetr gene?? Which proteins are you referring to

      Each gene is responsible for a particular protein production in all organisms. So here we insert our gene of own interest into the plasmid (pBR322). That gene can undergo the transcription and translation process and it can produce the protein. In that case if we use the chloramphenicol it restrict the synthesis of protein and the plasmid which carries our gene of own interest can undergo the replication and it won't produce the protein. That's why the copy number can be 1000-3000 per cell. If it produces the protein the copy number is controlled by the rop gene to 40.
