  • Mr. Andersen shows you how to sink leaf chads in preparation for the AP Biology photosynthesis lab. An empty syringe is used to remove gas from the leaves before the lab. As the chloroplasts absorb light they produce oxygen bubbles which eventually cause the leaves to float.

      @@WeAreBlank18 It's not really stealing. It's an incredibly common experiment and one of the easiest ways of measuring rate of photosynthesis. So common in fact, that it has its own measuring unit called ET50.

    My entire class got them to sink, but no one got them to float. We unanimously decided to pretend it never happened.

    I was forced to watch this as part of our pre-lab for our own Photosynthesis experiment...
    I am really tired as I procrastinated, and I just wanted to get it over with _(side note: it was easy as took like 15 minutes)_
    I watched this vid expecting to be bored, but I actually found it really interesting. Hope I do good at the lab tomorrow.

    Thank you kind sir, I have been stuck with a college bio lab at the last week before the semester since the person that had agreed to do it backed out. I have taught the lecture for several years, but this is new and I am scrambling and can't find a supply of rennet or rennilase for the lab I was going to do, so this will be a fabulous replacement integrating the sci method testing different wattages or distances from the heat sink.

    Summary of video: if you put light over plant cells, they can see the camera you are recording with. So, they fly away from the view out of embarrassment.

    Thanks for the video. Me and my teammates are looking for an experiment about photosynthesis and we're so out of ideas on how are we gonna put an experiment of it. Then my friend saw this video and it really helps... Now we're gonna use this for our experiment... The chads floating is fantastic... I like it :)

    the photosynthesis occurs in the leaf chad and they produce O2 in the leaf cavity, increasing their buoyancy, thus rising it up

    Would anyone know the purpose of pushing on the syringe after you have pulling on it? We did this lab a couple of weeks ago and nobody in our class was able to get the leaf chads to rise after sinking them in the syringe. I was surprised at how fast those leaves sank for Mr. Anderson as when I was duplicating the experiment, I spent upwards of 10-12 min creating the vacuum in the syringe to sink even a few chads.

    Can someone explain to me how the leaves generate oxygen bubbles without carbon dioxide? Mr. Anderson said all gas is removed from the leaf during the syringe step, so that would mean CO2 got removed too. Is there still CO2 in the leaf chads? Or is the oxygen producing step not reliant on CO2?
    Edit: I figured it out but decided to leave the comment here for anyone else who had the same thought.
    The light dependent reactions of photosynthesis are the ones that make O2 and the only input is air and light. So yeah. The step that needs CO2 is the Calvin cycle.

    I understand that the rate of photosynthesis is the independent variable and ET50 is the dependent variable. But I'm unsure of how to determine the rate of photosynthesis of ET50. Please help!

    Hi Mr. Anderson- My class did this lab, and I'm having trouble determining the dependent & independent variables, and the control. I assumed that the dependent variable was the number of chads, the independent variable was the amount of 02 in the chads when exposed to sunlight, and that the control was the amount of liquid in each cup. Is this correct? Thanks again for making these videos, they make bio so much more understandable!

      9 years later, maybe for another student... This is not an experiment, it is actually the control. You have to compare this with another condition, which is your independent variable (amount of light, color of the light, temperature, etc). Then what you measure (the time at leaves start to float) is your dependent variable (which is proportional to the amount of O2 or photosynthesis rate).
      Sorry if my english is wrong.

    Great video! It helps but you are a few labs behind us. It seems that I turn in a Lab Report a week before your video on that lab comes out! We just finished the Mitosis and Meiosis Lab in my class
    Of course no need to change the way you do things for one class.

    You don’t have to make a heat sink because we are using LED bulbs. But this is basically how you set it up.

      +Calypso Calypso Photosynthesis. The process that happens in the syringe pulls the air out of the leaf chads, then by creating a vacuum with the solution, it causes the Carbon Dioxide in the solution to be sucked in, replacing the air. When placed in the sodium bicarbonate under a lamp, photosynthesis starts and releases the byproduct of Oxygen, which increases the buoyancy of the chad and makes it float. We just did this lab in my class.
