  • Rats are also useful for aging research and for cooking ratatouille. But in all seriousness, take a look at this recent headline article - “We have the oldest living female Sprague Dawley rat,” said Dr Harold Katcher, a former biology professor at the University of Maryland, now chief scientific officer at Yuvan Research, a California-based startup.
    So, Rejuvenation & rats. That’s what we’re talking about today, and how this rat has apparently become the longest living rat for its species following concentrated plasma injections from young blood plasma, and what this could mean for human therapeutics, along my perspectives. But, before we get there we must go back, back to the late 1950s and early 1960...to a time when The Sheekey Science Show did not exist, but when researchers, such as Clive McKay did, and these researchers were conducting a procedure called heterochronic parabiosis.

      In one of his videos, Dr. Katcher speaks out about the '5' in his E5 compound and he simply states '... because 5 is our lucky number'. That seems to be it. So far.

    Just a minor point - the plasma is not from young mice. IT is from young piglets. In other words, the plasma is from a different species. I note that there are other experiments that show that plasma from young animals of one species has rejuvenating effect (when injected) on older animals of another species.

      Dog trials, which address this problem have already started, with human trials hopefully starting next year. It blows my mind that this gets far less attention and funding than many other longevity companies, but has the most immediate potential. Thankfully, it does not need a lot of funding.

      @@squamish4244 I'm not bullish on E5. A good chance that Dr. Harold Katcher will run out of time before E5 is proven to work. That is, unless it works so well that E5 extends Dr. Katcher's life by 30 years and he gets to finish his research.

      @@davidchung1697 Why not? I mean, I know he is 78 years old and so of course he's running up against the clock, as he is certainly well aware, and I know many interventions in rats do not work in humans, but why are you skeptical, specifically?
      In thirty years, society will be very different in many ways compared to today - certainly nothing like comparing 1993 to now - including in medicine, so predicting exactly how long it will take for any sort of longevity research to work that far out is a fool's errand.

    Really excited about these results! More than a treatment I think of it as a platform that is still unexplored. The ammount of things we could add to this blood extract is infinite so I'm sure we will have even greater news in the future.

    Great job on this video Sheekey!
    I read through the patent in some detail, and I was actually surprised that there are pretty specific instructions on how to make E5. Specifically, sections 191 through 199 describe in detail how to collect, process and prepare mammal/pig’s blood to produce E5, and sections 200 through 215 describe specifically how to administer it to rats for experimentation. The patent also appears to call out three specific tetraspanins as the apparent “active ingredients” of E5 (CD9, CD63 and CD81).
    I’d love to see someone try to replicate the results, but so far this whole thing appears to be mostly under the radar.

      It's all very weird and very mysterious. It almost seems like the plot of a mad scientist movie. I'll believe it when I see evidence outside of rats and mice - and like everyone else, I am SO tired of 20 years of headlines about amazing results in...rats and mice.
      When they start getting good results in dogs, then, and only then, will I truly start to be impressed.

    Sheekey it occurred to me the lab rats are genetically selected to not have much variation in their genes to confound the results. However ultimately experiments will need to be conducted on "rats in the wild" rats with variation in their genes. If we want robust therapies for the ageing process for humans all over the world gene variation will be a factor in it's effectiveness.

    May I suggest possible meaning for the name E5? E is obviously for Elixir, ("a magical liquid that can cure illness or extend life"); The Roman numeral for 5 is V. E5 means Elixir Vitae (Elixir of Life).

      5 was chosen because it was proposed as a lucky number by one of their colleagues. But I like your take on it.

    So far, most of the proprietary lifespan/healthspan improving supplements have been used to improve the health of the developers bank accounts. We'll see if this time is different, but even if it's not, at least we got some really old rats!

      Katchner claims that E5 can be produced from industrial waste so inexpensively that nobody on earth will not be able to have it. As the blind man said, "We shall see."

      It can't help but be inspiring that an 80-year-old man is doing this work, even though he is open about the fact that he may never benefit from it. That lends it more credulity to me, as many longevity researchers (inevitably - I'm not judging) are hoping that they will succeed in time for themselves, which may distort their judgement.
      Harold is also not actually a late-bloomer. He co-discovered the BRAC1 breast cancer mutation in 1994. His credentials and accomplishments are impeccable.

    Thank you! Wish this was open source. Apparently, the patent is very opaque. I still think there is a lot of potential with plasma dilution which can be done free (with smaller but significant volumes) even though some are trying to monetize it as well (ref. Lyspan launch - Kiprov and Verdin (of Buck) etc). A 2022 Conboy paper provides human data and useful biomarkers, some of which can be easily bought and tested for by anyone who wants to experiment with plasma donation.

      @@bw4611 Thanks, I'll see what the literature says about these. I'm a bit surprised Katcher isn't testing on himself considering his age and strongly he feels about the efficacy of E5. He could assess himself before and after using the same markers he's used in the rats plus do before and after proteome and other accepted tests including full medical workups. If he could show good results for himself he'd have plenty of funding to massively scale up production and accelerate more testing.

      @@peterz53 I agree. Interestingly, he did do an external test on the back of one of his hands, and the results were pretty noticable. But take with a grain of salt, as we only have comparisons of one hand to the other -- not a before and after.     Experiments require independent replication. Unclear if these were double blinded placebo controlled experiments. Also of concern would be results from a single source, i.e, Indian laboratories. I

    I confess, I need to pay more attention to this video. But is there any takeaway that platelet or plasma donation may have rejuvenating properties?

      I think there is a good case to made for regular plasma donation (~25% of plasma volume every 4 weeks) which can be done in US at conventional blood banks. Conboy Lab 2020 and 2022 papers show major changes in proteome and some tissues. Over the course of 2022 I donated about 3.5 plasma volumes (11 liters) with a combination of plasma donations intermixed with platelet donations. While I did not see any obvious changes, at 68 yo I have not noticed any changes in grey hair or skin. my liver function did improve and I have very good blood work overall This year I will be tracking some of the biomarkers mentioned in the 2022 Conboy paper (DNA damage etc). You might look up the 2021 Plasma Dilution paper by Rob Flickenger which appeared to show that normal plasma donation (about 800 mls) caused many of the same proteome changes observed by Conboy as shown in their 2020 paper.

    It was my understanding that it is called E5 because it has 5 ingredients, but I cannot find the article where I read that. And yes originally it was called Elixir.

      There is also a Dr. Sinclair's video when he first knew about Dr. Katcher's E5, were he disrespectfully says that 'how can anything serious can be expected from a compound called Elixir?'.

      Katcher said the number was suggested by a colleague, who mentioned that it's considered to be a fortuitous number. There is no deeper meaning behind it than that.

    Dear Sheekey if you read this comment please consider researching the connection between orgasms/having sex and aging. The goal of the organism is to reproduce and have its offspring reproduce. I think that longevity genes exist to increase an organism’s chance to reproduce. Sex doesn’t occur as much during times of danger so longevity genes turn on to give the organism extra time to have a chance at reproduction? If an organism isn’t having sex, wouldn’t it make sense for longevity mechanisms to become activated? I can’t find any research on this topic. Perhaps in our evolutionary past lack of sex coincided with starvation so there was never an evolutionary pressure to evolve longevity genes that get activated directly by lack of sex. Or perhaps my view is flawed somehow? If someone has any ideas or information to share on this topic, please do.

      @@austinknows456 When people think they might die, they want to experience life more. You see this in people during wartime. There were legendary amounts of sex during the world wars, and all the combatant countries handed out massive amounts of condoms. Stalin even asked Churchill specifically for condoms because the Red Army had such a shortage.

    Bad Science.
    The oldest rat ever on record was a pet named Rodney, who lived to be seven years and four months old, according to Guinness World Records. Rodney was born in January 1983 and died in May 1990. Rodney lived in Tulsa, Oklahoma, and had the same name as his owner Rodney Mitchell.

      @@Freja_Solstheim Doesn't invalidate my argument. Do Spague-Dawley rats have a disposition to die young because of a genetic condition that is ameliorated by the E5 plasma? When pet rats with good care can outlive lab rats by such a large amount then what does that tell us? Does the e5 plasma cause lab rats to chill out and live longer?

If lab rats by default only live 70% the lifespan of pet rats then how does this experiment prove that the extension of life is due to a longevity factor and not a reduction in the factors that shorten the lifespan of lab rats or due to the overcoming of the genetic disposition of these breed of rats.

      Lab rats are completely different type of rat from the pet rat Rodney. You can’t compare the age of a lab rat to a different pet species and claim that it lived longer.
