This example is grammatically correct, but it doesn't make much sense. Every person has a birthday. The better, more natural question is a Wh- question such as "When is George Washington's birthday?" But good try!

We can't say "Yes, I'm / Yes, you're / Yes, he's / etc." I'm not 100% sure why we can't, but my guess is that it's difficult to hear a contraction in a response because it's so short. It's interesting, though, because we CAN do this with negative forms, but maybe this is because they're a bit longer and easier to hear. For example, it's not correct to say "Yes, you're" but it is correct to say "No, you aren't" (or "No, you're not").
