They're already assigned instrumets from the start. If you actually listen to character soundtracks Garnet's is a synth. It always has been a synth. And Amethyst's aren't solely based on drums, but they have a lot in them because that's what she plays. That's why Sugilites has heavy toms, a tam-tam or gong and synth.

In autoplay mode, instruments that I don't have plugged in won't count towards score, look at 8:22 for example, the score isn't going up at all.
