High-throughput monitoring of cancer cell/fibroblast co-cultures: interaction with TNF-α

  • The effect of both the seeding ratio and TNF-α concentration on proliferation of HeLa cells (cervical carcinoma cell line; green) and 3T3 cells (fibroblasts; brightfield) in co-culture was investigated using the CytoSMART Omni FL and corresponding Cloud-based image analysis algorithm for brightfield and fluorescent channel confluence. The kinetic confluence profiles of cultures with three different seeding ratios - 1:0, 1:1 and 0:1 (HeLa : 3T3) - and three different TNF-α concentrations - 0 ng/ml, 2 ng/ml, and 20 ng/ml - were determined.
    This experiment revealed competition between HeLa cells and 3T3 fibroblasts in co-culture. Higher concentrations of TNF-α suppressed cancer cell survival and promoted fibroblast growth in co-culture, whereas this effect was opposite in mono-cultures.
