  • Macroautophagy: Steps, Regulation, and Importance in Health
    Hey guys! In this lesson, you will learn about the macroautophagy pathway, how the pathway is initiated, how the pathway operates, what regulates the pathway and why the pathway is important in human health. You will also learn how different types of pharmaceutical drugs act to inhibit or activate the macroautophagy pathway.

      FYI, guys, Dr. Berry has his own RUclips channel where he translates concepts like this into practical steps you can implement to improve your own health. I don’t always agree with him (especially on the subject of iodine intake), but he is very informative like JJ here - just not as technical.

      Yeah, agreed. I tend to think Autophagy for dummies is simply; ‘fast long enough for autophagy to start, smile, wait 4 hours, eat, repeat’ and leads to OMAD, fasting, and regularly eating 16:8.

      My understanding of all that is if you’re eating a diet that does not include sugar and grains. And you fast 16to 20 hrs or 36 hours it triggers Autophagy. Correct me if I’m wrong.

    I have a question. You say that TFEB is a transcription factor leading to the transcription of ULK1, BECLIn1... which you mentioned as Lysosome Biogenesis. But in this daigram it looks more like Autohagosome biogenesis. How are these proteins also contributing to lysosome biogenesis?

      Listen to Dr. Jason Fung and Dr. Nadir Ali. I If daily at 21/3 or 22/2. I have heard it begins at 18 hours. How accurate that is is questionable. You raise a valid question and one I have asked myself many times. I fast as well. Last fast I did was a 5-day pure water and electrolyte fast.

    No, the Vps34Beclin1Vps15 complex does NOT convert PIP2 to PIP3. That is the activity of Type I PI-3 Kinase, which is NOT required for autophagy. Vp34 is a type III PI-3 Kinase that converts PI to PI3P, phosphatidylinositol-3-phosphate. It is essential for autophagy, especially the fusion of autophagosomes with lysosomes.

    wow nice explanation. i have question can autophagy fight viruses like hsv ect?

    • @jjmedicine
      @jjmedicine  4 года назад

      Hey Aldi! Thanks for the comment! Yes, autophagy is known to fight viruses by way of "virophagy" (they can engulf and degrade them like they can with some bacteria). However, some viruses have found a way to use virophagy for their own purposes so that they can infect cells. Unfortunately, it does seem that HSV is one type of virus that can use autophagy to help propagate itself.
      Hope that helps :)

    Hey, are you sure that VPS34 converts PIP2 to PIP3? VPS34 is a class III PI3K, so it actually should convert phoshpoinositol to PI3P rather than PIP2 to PIP3, right?
