Soxhlet apparatus is one of the most important apparatus used in Pharmacognosy and botany practical class. Soxhlet apparatus is used for the extraction of crude drug for evaluation of its phytochemical and medicinal importance. Soxhlet apparatus is made up of glass assembly of borosilicate type. Soxhlet apparatus consists of 04 main parts or section-
(1) Round bottom flask
(2) Drug chamber with vapour and syphon tube
(3) Condenser unit
(4) Water pipe.
Soxhlet apparatus is also known as a hot extractor of crude drugs. The drug which is to be extracted is firstly converted into coarse powder and then filled into a thimble. In Soxhlet apparatus thimble is a pocket made up of either filter paper or muslin cloth, which allows only solvent to pass from and it does not allow drug particle to pass with the solvent used. After adding the drug to crude drug chamber the appropriate solvent is introduced through the solvent inlet over the condenser. this solvent comes down at the round bottom flask, a heat by hearting device coverts solvent into vapour and this vapour travels through vapour tube and at the bottom of the condenser in Soxhlet apparatus it condensed and converts into the solvent and again fall down over the crude drug and the level of solvent is increased. When it increases in sufficient quantity that comes down to the round bottom flask via syphon principle and this steps continues till solvent becomes concentrated. This ends the process of extraction.
This video will give you knowledge about- Soxhlet apparatus, extraction process, heat extraction, parts and function of Soxhlet extraction assembly.

Excellent explanation. mam, I have just one question. if I take 5g crude, then how much volume of solvent is required for extraction, and what will the duration time for heating?
Where is the part two of the video wherein you evaporate the solvent and get the concentrate?
Thank you soooo much madam you SAVED meee ❤🥺 bahot shukrya madam ji
Very detailed video! The other sample though was not completely soaked prior the siphoning. This may result in less efficient extraction.

What should we do when the solvent in the round bottomed flask dries up
Excellent demonstration
Nice explanation
Hi is it better to get a 250, 500 or 1000ml round bottom flask?
Thanks mam from Bangladesh.
Great and informative video.! But is there any way by which one can minimize wastage of precious water by somehow circulating the same water through the condenser.?
lamniang tombing Yes. We did the same. You may collect the outlet water in a bucket and then place the inlet of condensor thus the cycle will continue without loss of water.
YOU HAVE TO USE A BUCKET AND A WATER PUMP, PLACE BOTH THE OUTLET AND INLET IN THE SAME BUCKET THEN THE CYCLE WILL CONTINUE WITHOUT ANY LOSS.
Rishabh Kaundal yes. We follow the same. But to explain better and for video purpose we have not used bucket.
Thank you for this video. Your explanation is very detailed and its made something I thought was difficult seem so doable. I leave this video feeling so confident . Thank you!!
Excellent video

Can I use propylene glycol as a solvent to extract tinctures from dehydrated fruits, cocoa and tobacco?
You should not use the propylene glycol as an extraction solvent, as you need something with a lower boiling point and viscosity to easily penetrate the material for percolation. Its higher viscosity and high boiling point does however work well for making a tincture out of your extract. Once you have extracted the desired compounds with your volatile solvent, you could evaporate it off and disolve in propylene glycol or a mixture of propylene glycol and glycerin to make a tincture to use as an e-liquid for example. For the plants listed I would recommend the following:
Cocoa - Use an aqueous extraction; repeat 3x to ensure maximum recovery. Concentrate it to a third of its volume,
add sodium carbonate to the aqueous phase and extract this using a nonpolar solvent 3x. Evaporate/distill
the nonpolar solvent to obtain crude theobromine. From here you could recrystalize using an ethanol/water
mixture to purify and/or make a tincture with glycerin or oil of some sort.
Tobacco - With tobacco, you would extract via percolation with a polar solvent. It is highly water soluble so just
repeat a couple times with basified hot water and evaporate down to obtain a crude residue of tannin salts
and nicotine. Disolve the nicotine out of the residue using some hot acetone or something like that, and
then boil that off and disolve in glycerin or propylene glycol to make an e-liquid of sorts.
Dry fruits - That depends on the alkaloid or tannin profile. Most of the time just go for acetone or ethanol, but you
may need to use the water/nonpolar solvent method on these sometimes.
Hope that helps you. Just remeber to check wikipedia or PubChem for the solubility profile of whatever the specific thing you're aiming at prior to chosing a solvent, but for the most part viscous, high boiling solvents are a terrible idea and are only good for tinctures and e-liquids.
@@williamackerson_chemist Thank you so much for taking your time to answer my question. I really appreciate your help. Cheers!
