We use the Bunsen burner, since we consider an aseptic area, this is very important when you are working with bacteria (all microorganisms btw) but ... why don't you do it like that? : /
Might just be our protocol/SOP, but anytime we are working with competent cells we are using sterile technique, in the sterile field around a Bunsen burner. Is that just not your labs standard protocol?

dont you use a selection marker during your transform?
kwinchee yes, but it’s still just our protocol to attempt to avoid contamination or any other issues.
