It discusses Ion Selective Electrode , its principle and live demonstration of serum electrolyte analysis.
A.) ISE is a more accurate and specific method.
This works on the principle of a potential created by the difference in concentrations of electrolytes on each side of a selectively permeable membrane. In this system two electrodes are used where the reference electrode has constant potential.
The ion selective electrode develops a voltage that varies with sample concentration. Thus, the potentials difference between the reference electrode and the ISE, is calculated and expressed by Nernst Equation. and displayed in mmol/L or mEq/L on the digital displayer
B)Flame Photometry It is one of the oldest direct potentiometric methods commonly used to measure the concentrations electrolytes in serum urine samples.
Principle:The flame photometry principle is based on chemical property of alkaline‐earth metals on high temperatures absorbed energy from heat source being conducted to state of excitation in their atomic form.
When they return to initial energy state the radiation is release at specific wavelengths being some in region of the visible spectrum.
The emission is proportional to concentration of ions present at the sample. A photodetector behind the filter gives an electrical signal which is proportional to concentration of electrolyte in the sample.
In most routine diagnostic laboratories, ISE is method of choice for Na+ and K+ assay because it is:
reliable,
selective,
sensitive and
small sample volume is required.

What is roll or mechanism of reagent in machine, plz explaine
The reagent pack fir an ISE electrolyte analyser has, serum standards, diluents, and reference standards. The concentration of electrolytes is compared to that of internal standards. When compared to the Internal Reference Solution, this electrical potential is translated into voltage and then into the ion concentration of the sample.
