You’ll only want to dip the toothpick once into the agar stab for the first streak. After that, you’ll want to drag the second toothpick through the area that they first swiped to pick up a small amount of bacteria.
Thanks for the question! You should use sterile technique when working with media containing antibiotics. Depending on the contaminant, it may be resistant to the antibiotic in your agar.
To help maintain sterility. If you're working with a wire loop, you will sterilize the loop in the flame, cool down by touching the agar plate on an area that does not contain bacteria, and then use to streak

Do we need to dip in bacteria culture for every single toothpick or just for the very first toothpick?

Aren't there any devices rather than a flame? Such as device made with UV bulb or so? If such present, will that be effective?

As far as I'm aware gloves should not be worn when working next to the flame
