The narrative does not match the video. It’s not a 1:1 ratio of blood to PBS; from what I observe, they use at least three volumes of PBS to dilute the blood.
Correct me if I'm wrong.

Phosphate buffered saline. It is a buffer at pH similar to blood so that the cells are happy and don't suffer osmotic shock.
