I think it all depends on the finances as to whether you use silk or cotton.

I want thread to fill the square. Ive used DMC and find that the black thread is horrible and some other colors as well. Im even going to use 16 count to see if that will cover better.

I am trying to figure that out too ! I think it might be 3 strands as it looks like one strand was cut
