      no, they do have. if you imagine that they have 2 strands so at any side there would be 5prime phosphate and a 3 prime hydroxyl.....so what people do is phosphorylate the hydroxyl groups in those position so all are now phosphate group...and the vector they do opposite they make all hydroxyl group by Alkaline phosphatase.This method ensures a better ligation
