I use 1ml column to purify protein from e.coli cell lysate, but I always get multiple bands after a check on the western blot. I have tried to increase the concentration of Immidazole on the binding buffer (up tp 50mM), and 500mM for elution buffer, but still faced the same problem. Do you have any suggestions to solve this problem? thank you
