Why, if the restriction enzymes are located on either side of the segment being removed just like in the large section, is the the exonuclease needed and why do you end up with 2 of the small segments on each end of your plasmid once it's been cleaved. Are the restriction enzymes cleaving directly down the middle of the segment to be deleted?
