For example different templates, single page but different design if that makes sense. Is it possible without making a subdomain with each installation of wordpress and theme etc?

It would depend on the specific autoresponder you are looking to use but some of the others do connect to APIs. We'll keep an eye out but if you reach out to the plugins directly then can give you a better idea of when they plan to add an API to their service.

While possible, that would be a complicated setup and we currently do not have a beginner friendly method for setting that up.

It heavily depends on your audience and content. Without testing it is hard to say which would work better for you.
