I don't want to go through heavy spoilers, but essentially, our antagonists with guns formed a contract with the gun devil, who has told them that they want denji's heart. They assassinate the devil hunters since they are the only threat to them achieving that goal. Katana man has a personal grudge against denji since his grandpa was the mafia boss that denji killed in episode 1. As for why they want his heart, lets just say that Denji/Chainsaw Man has a very unique ability that all devils fear, so the gun devil wants his heart so it can consume it and prevent him from reviving.
