  • Maria has been living with HIV for more than thirty years and is an ambassador for National Women and Girls HIV/AIDS Awareness Day, a blogger, an HIV advocate and activist who is dedicated to building awareness about HIV and working to reduce HIV stigma. Maria is also a global ambassador for a non-profit women’s organization called The Well Project, which is working to change the course of the HIV/AIDS pandemic with a comprehensive focus on women and girls.
    About The Cure Chronicles:
There are more than 38 million people in the world currently living with HIV/AIDS. At American Gene Technologies® (AGT™), we believe there is hope for a cure. But it will take everyone working together to make it a reality and permanently end the suffering of millions of people living with HIV today.
AGT is testing a single-infusion cell therapy (AGT103-T) to hopefully cure people living with HIV. Meanwhile, we want to spotlight some of the many people in the HIV community who strive to keep hope alive and tip the odds toward success.
The Cure Chronicles video series features thoughtful conversations with people living with HIV, advocates, medical experts, policymakers, and others working hard toward the shared goal of ending the devastating HIV epidemic.

We are estimated to have a cure by 2030, possibly a little bit earlier than that depending on the progress.

      Excision BioTherapeutics' EBT-101
      or
      American Gene Technologies' AGT103-T?
Which possible HIV "functional" and/or "sterilizing" therapeutic cure will be the first to come out of human clinical trials and FDA approved for safe usage?
[1]. Excision BioTherapeutics' *EBT-101* as a potential "sterilizing cure". EBT-101 is currently in human clinical trials and in September 2022, Excision BioTherapeutics dosed their first participant in the EBT-101 Phase 1/2 Trial. EBT-101 is CRISPR-based, single administered therapy, designed to cut and removing HIV DNA out of human cells. This potential gene editing treatment employs an adeno-associated virus to deliver CRISPR-CAS9 nucleases and dual guide RNAs targeting three sites within the HIV genome that tell the enzymes where to cut. The
website reference and for additional information on Excision BioTherapeutics and EBT-101, please go to
      www.excision.bio/ , as well as clinicaltrials.gov/ct2/show/NCT05144386 .
[2]. American Gene Technologies' *AGT103-T* as a possible "functional" cure. AGT103-T is currently in human clinical trials and in June 2021, the first patent was treated with potentially curative HIV cell and gene therapy. AGT103-T is a cell product made from an individual's own cells and modified with AGT103 lentiviral vector for HIV resistance. The Phase 1 trial found that AGT103-T was well tolerated in humans and persisted in the body. AGT103-T is designed to restore the Gag-speciffic CD4+ T cell response in persons with chronic HIV who are receiving antiretroviral therapy. The website reference and for additional on American Gene Technologies and AGT103-T, please go to www.americangene.com/pipeline/hiv-aids/
There is also the possibility of stem cell transplants from donors with the *CCR5Δ32 gene mutation* , as "sterilizing" HIV cure.
On February 20, 2023, a 53-year old man in Germany, named the "Dusseldorf Patient" (Georgia native, Paul Edmonds, of the United States, that later relocated to San Francisco) has been documented as the *5th person totally cured of HIV* by receiving a stem cell from a doner with a *CCR5Δ32 mutation* , which blocks HIV from infecting the body. The 53-year old patient was diagnosed with HIV in 2008, and after their HIV diagnosis, was placed on antiretroviral therapy (ART) which suppressed the viral load within their system. The patient enrolled in the University Hospital Dusseldorf IciStem program, which explores potential HIV cures requiring stem cell transplants. This 53-year old patient was diagnosed with acute myeloid leukemia about three years after his HIV diagnosis. Remission of the cancer was achieved through initial rounds of chemotherapy, but was followed by a relapse shortly after.
During the 53-year old patient's treatment of his cancer, he received a stem cell transplant from a female donor with whom they matched, and the female donor had the CCR5Δ32 mutation. This stem cell transplant occurred about two years after the 53-year old patient's cancer diagnosis and five years after being diagnosed with HIV.
This stem cell transplantation, was a person with the CCR5Δ32 mutation is a complicated procedure, normally reserved for cancer patients, and comes with many risks, and is too risky to offer it as a option as an HIV cure for everyone.
Some individuals were born with the CCR5Δ32 mutation, which naturally blocks HIV, there were those that are considered LTP (Long-term Progressors), which they will carry HIV for decades before their HIV progress to AIDS (with HIV medications).
