That's a very good question. I have found varying %s of glutamate conversion. I am listing 1 reference below that mentions 52%. Just remember that polyglutamate residues do not leave the cell, but has the activity of MTX. You can read the below article. Kuehl M, Brixner DI, Broom AD, Avery TL, Blakley RL. Cytotoxicity, uptake, polyglutamate formation, and antileukemic effects of 8-deaza analogues of methotrexate and aminopterin in mice. Cancer Res. 1988 Mar 15;48(6):1481-8. PMID: 2449950.
