    Colorimetry
    In this technique , the concentration of compound is determined by measuring the intensity of color.
    Colorimeter
    They are simple instrument which measure the intensity of transmitted light through color solution
    Beer’s law
    When a parallel beam of monochromatic light is passed through a solution, the absorbance (A) of the solution is directly proportional to concentration (C) of the substance in solution
    A = KC
    K is a proportionality constant
    Lambert’s Law
    When a parallel beam of monochromatic light is passed through a solution, the absorbance (A) of the solution is directly proportional to length of the light path (l)
    A=Kl

    Sir good afternoon.sir I have doubt. That I am using Beacon brand colorimeter for biochemistry test.doubt is that.my colorimeter OD range is 2.00 to -0.31. I 1st gave blank and set zero,after remove blank the OD is 1.69.is it correct working my colorimeter.

      1st you have to set the zero and then you have to measure the od of blank

      Take the OD of blank standard and test and enter the value in formula and calculate the total amount of sugar present in the solution

    Sir why we set the reading at zero after inserting the cuvette?

      Zeroing a colorimeter before taking a reading is important because it allows for any background color or light to be accounted for and removed from the final measurement.
      When a colorimeter is zeroed, it essentially establishes a baseline reading of the blank or clear solution, without any sample present. This baseline reading accounts for any color or light that is naturally present in the system, such as ambient light, impurities in the reagents, or any residual color from a previous sample. By zeroing the colorimeter, these potential sources of error are accounted for and eliminated from the final measurement.
      Once the colorimeter is zeroed, the sample can then be added and its absorbance or transmittance can be accurately measured. This ensures that the final reading is a true representation of the color or concentration of the sample, without any interference from background color or light.

      A complementary filter allows only the color of light that the solution absorbs most to pass through. By eliminating other wavelengths, the colorimeter focuses on the interaction between the light and the colored molecule.

      I need only a small quantity preparation of working reagent to per patient test.From the kit insert,Working reagent must be prepared by adding all the enzyme reagent bottle(3ml) to Buffer reagent(60ml).and it is stable for 16 weeks.My testing load is very low.I need to prepare working reagent per patient.But I don't know the proportion of mixing.
