GraphPad isn't letting me chose Tukey's method. It only lists Dunnett's T3 test and some others that are not Tukey's method. Is there a way to force the software to let you chose Tukey when it doesn't list it as a recommended test in the options tab?
You first have to plot a standard curve, that is measure the absorbance of standard of known concentration at different dilution levels. Make sure your data is all baseline corrected (subtract a blank). Then plot a linear regression of absorbance as a function of concentration. Once you have the equation for the linear regression, you can plug in the absorbance of your unknown and use algebra to solve for the unknown concertation.
