The only "variation" that could arguably be considered official is replacing arigatou with arigatō. But that's retarded. ō is not even a thing in either english or japanese. There's absolutely no reason for it to exist. Where does it even come from? Sometimes it replaces OU, sometimes it replaces OO. WHY? You literally have established that お is "O" and う is "U", but then you tell me that sometimes either おお or おう is replaced by some other thing that doesn't exist in either language. Many Japanese words look like Chinese words, some of them are almost same...
It's because Japanese use the Kanji from the Chinese and either use the Chinese's original meaning of the word or they use their own.
