Sir, in benzedine test- for heamoglobin , if WBC is present result will positive wrongly ,so we do heat , WBC is destroy but why RBC is not destroy at the time?
H2O2 is broken down to nascent oxygen by heme present in hb, the nascent oxygen is the main component for conversion of benzidine into blue or green unstable complex which indicates presence of blood in urine
To determine if the coagulum formed is by protein or by jon protein substance (Phosphates or carbonates).. If it is by protein then the coagulum won't get dissolved in glacial acetic acid

Rbc has hemoglobin which is heat stable thats why it doesn't get destroyed
