If you are using the anni ship, there does seem to be a small chance to fail the 5* while letting it auto its way through. The seems to be a combination of having less HP than when using the Whale Sharks ship, potentially receiving the bleed debuff on stage 4 and the bot hitting quick taps.
Will need to replace more than just her. Replace Ulti with legend Smoothie and swap her with the FC, take Queen off as a sub and put him as a support on King, and fill the empty spot with either TM RR Urouge or Legend Chopper that has Support RR Brûlée on him as a support. Lastly, you'll need to take off V1 RR Robin support and put on V1 RR Nami support on Reiju and Raid Shiki support on Bartolomeo Cavendish. The damage is cutting it a little close, but should be able to clear. Extra cotton candy could help. If you use Urouge, then you'll only be down one more booster; Otherwise, you'll be down 2 boosters if you use Chopper. For the lower tier difficulty, you should be able to replace her with any other booster.
You could also try 6+ Roger as your FC instead of Smoothie. I'm unable to input this into the damage calc at the moment, but with everything else, I'd think it would get past stage 4 jut fine. You'll use Roger on stage 5, so you won't need the Raid Shiki or V1 RR Nami support. This means you could also use M56 Nami or RR Anni Nami as supports on Chopper if you don't have Brûlée.Only downside in all of this is that your FC Roger won't hit for much.

Do you think it would still work with 6* only barto/cav ? I didn't get the last skull in the last kizuna
You'll need to replace them with someone else, since the 6* version won't be able to reduce the enemy's threshold damage reduction.
